The words will be counted each time you upload a content to the Content Storage, this will refer to the words that the content has when generating its full text. They will also be counted when recreating or editing the generated chunks.

The words of content spent are counted directly in the limit of your Promptitude plan, remember that they aren't renewable (similar to your number of prompts), it is a unique amount.
